Development of molecular marker for mutagenic detection due to heavy metal in lettuce
Funding period : 2011- Deactivate
Abstrak
This study aims to evaluate the mutagenic effects of heavy metals based on changes in DNA patterns with PCR-RAPD and PCR-ISSR. The method used is planting in a hydroponic system, by administering concentrations of Cd and Ni separately at concentrations of 5 ppm, 10 ppm, 20 ppm and 30 ppm. The treatment is given after the plants are 5 weeks old in the medium ΒΌ MS. The medium is changed every week. DNA extraction to test the Cd and Ni genotoxic effects was carried out from leaf samples according to the Doyle and Doyle methods. PCR-RAPD was carried out using 8 RAPD primers, while PCR-ISSR used 9 primers. Polymorphisms were only obtained in the treatments of heavy metals with high concentrations
