Comparative study on physicochemical and biological characteristic of collagen extract by acid and hydrothermal method from swim bladder of yellow-pike conger (Muraenesox talabon)

Indonesian capture fisheries production was abundant and consequently the fisheries processing industries resulted huge fishery waste. One of the waste was swim bladder that high in collagen, XXX potential be developed as biomaterial, cosmetics, pharmaceutical supplement. Yellow-pike conger (Muraenesox talabon) was one of by-catch fish which frequently got in every tuna catching. This aim of this study was to determine and compare the physicochemical and biological characteristics of yellow-pike conger swim bladder which extracted with different methods. Different extraction methods affect the physicochemical and biological characteristics of swim bladder collagen. Acid and hydrothermal extraction method of collagen from swim bladder revealed the occurrence of α, β and γ bands with more than 70 mg/100 mg collagen corresponding to more than 70% of collagen and thus confirmed the nativity and purity of the swim bladder collagen. FT-IR studies confirmed the retention of all three amide bands of I, II and III, and triple helixcity. Swim bladder collagen matrix promoted high cell growth overlapping normal cell viability (without collagen) after treatment with 1200 ppm concentration. The optimal cell attachment started at 600 ppm collagen treatment and shown better proliferation with uniform distribution confluent.
