CVPD Disease in Citrus Plant - The New Biotechnology Based Analyses

Abstrak

The disease of CVPD in Citrus plant is also called citrus greening disease or huang long bing is the main disease of citrus.  The causative agent of this disease previously to be said was a virus based on the symptom of the infected leave of citrus plant.  Later on, it was found by the electron microscope that the disease was caused by mycoplasma.  That way the citrus plantation infected by the disease was treated by tetracycline, however, the treatment was failed to get good result.  Why ? Because, the causative agent then found was a BLO (Bacterium Like Organism).  It was also found that it has a thin cell wall consist of outer and inner membrane, like a Gram negative bacteria. Then in line with development of biotechnology, molecular identification testing was carried out with Polymerase Chain Reaction (PCR) to identify the causative agent of CVPD disease.  By using a 16S rDNA a conserve sequence in bacteria as a primer to amplify the DNA target sequence.  The amplified DNA fragment then analyses by using restriction enzymes it was then constructed the specific primer the bacteria that caused CVPD disease.  So it can be ascertained that the cause of CVPD disease in citrus plant is a Gram negative bacteria called Liberibacter asiaticus.


This book also contains the results of our research such as PCR identification of all varieties of citrus plants in Bali and surrounding island and research on in vitro culture of citrus plant in the making seedling free L. asiaticus.   Beside that, the book also contain the results of the research of isolation and molecular characterization of a DNA fragment that considered to play an important role in CVPD disease resistant. The molecular cloning in plasmid vector pUC 18 and pBI121 and the in vitro genetic transformation of CVPDr DNA fragment also explained in the book.
