CONSERVATION OF BALI FORMA DENDROBIUM THROUGH IN VITRO CULTURE

Abstrak

Dendrobium orchid is one of the orchid genus that has a high diversity of species, spread in several regions in Indonesia including Bali. The diversity of natural forma Bali Dendrobium orchids is a great potential as a source of germplasm for the development of Indonesian orchids, a means of learning, research, and other potential excavations. Based on the results of explorations that have been done, there are three potential species of Dendrobium forma Bali namely D. macrophylum A. Rich., D. secundum (BJ) Lindl and D. heterocarpum Wall.ex Lindl is threatened with extinction because the population is very small due to illegal hunting, damage forest habitat and conversion. Seeing this phenomenon conservation or conservation efforts are very urgent to do so that the wealth of this germplasm can still be maintained and developed.
In this research efforts will be made to propagate these three orchid plants in vitro through seed culture and organ culture. There are 3 stages of activities in this study, namely: 1) Dendrobium orchids in flowering are polled by selfing and sibling to obtain orchid pods containing seeds. 2) Sowing orchid seeds in vitro on a variety of media with the addition of organic matter. 3). Dendrobium orchid species that do not flower, propagated in vitro through organ culture on induction media with the addition of auxin ZPT and cytokinins. The second and third stage of the activity will produce propagules (explant material to be directed to produce young plantlets / plants in subsequent studies).
The research that will be carried out aims to: (1) increase the population of the endangered Bali Dendrobium forma orchid, (2) find media formulations for each type of culture method in vitro, (3) the results of this study are expected to contribute to the enrichment of science and technology, among others, through the publication of research results in international journals and disseminated at the ICoSTH international seminar.
