Pengaruh Pemberian Ekstrak Etanol Daun Ashitaba (Angelica keiskei) Terhadap Histopatologi Lambung Mencit (Mus musculus) Jantan

Abstrak
ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol daun Ashitaba (Angelica keiskei) terhadap histopatologi lambung mencit. Setelah diadaptasi selama satu minggu, mencit (Mus musculus) jantan yang berjumlah 25 ekor dibagi secara acak menjadi lima perlakuan, sehingga masing-masing perlakuan berjumlah 5 ekor. Perlakuan pertama tidak diberikan ekstrak etanol daun Ashitaba atau sebagai kontrol. Perlakuan kedua sampai keempat diberikan masing-masing dosis 125 mg/kg bb; 250 mg/kg bb; 500 mg/kg bb; dan 1.000 mg/kg bb dalam waktu 21 hari secara oral. Nekropsi untuk pengambilan organ lambung dilakukan pada hari ke 22. Jaringan lambung selanjutnya diproses untuk pembuatan preparat histopatologi dengan pewarnaan Hematoxylin Eosin (HE). Hasil dari pemberian ekstrak etanol daun Ashitaba (Angelica keiskei) dengan dosis 125 mg/kg bb, tiga ekor mencit mengalami degenerasi dan infiltrasi sel radang dan satu mencit mengalami nekrosis. Pemberian dosis 250 mg/kg bb, tiga ekor mencit mengalami degenerasi dan 2 ekor mengalami infiltrasi sel radang dan nekrosis. Pemberian dosis 500 mg/kg bb, tiga ekor mencit mengalami degenerasi, infiltrasi sel radang dan nekrosis. Pemberian dosis 1000 mg/kg bb, tiga ekor mencit mengalami degenerasi dan nekrosis, dan empat ekor mengalami infiltrasi sel radang. Dari hasil penelitian ini, dapat disimpulkan bahwa pemberian ekstrak etanol daun Ashitaba (Angelica keiskei) secara oral dengan rentang dosis 125 mg/kg bb sampai dengan dosis 1.000 mg/kg bb selama 21 hari tidak menimbulkan gangguan struktur histopatologi organ lambung mencit (Mus musculus) jantan. Kata kunci : Ashitaba, lambung, mencit ABSTRACT This study was intended to find out the effect of ethanol extract of Ashitaba leaves (Angelica keiskei) toward the gastric histopathology of male mice. After being adapted for one week, 25 male mice (Mus musculus) were divided randomly to be five treatments, so that in which each treatment consisted of 5 male mice (Mus musculus). The first treatment was not given ethanol extract of Ashitaba leaves which was called as control. The second till fourth treatment was given each dose of 125 mg/kg bb; 250 mg/kg bb; 500 mg/kg bb; dan 1.000 mg/kg bb during 21 days orally. Necropsy for gastric organ harvesting was conducted on the day of 22. Then, the gastric tissue was processed for making histopathology preparation with Hematoxylin Eosin (HE) staining. The result of giving ethanol extract of Ashitaba leaves (Angelica keiskei) with a dose of 125 mg/kg bb showed that three mice undergo degeneration and infiltration of inflammatory cells and one mice undergo necrosis. The dosing of 250 mg/kg bb showed that three mice undergo degeneration and two mice undergo infiltration of inflammatory cells and necrosis as well. The dosing of 500 mg/kg bb showed that three mice undergo degeneration, infiltration of inflammatory cells, and necrosis. The dosing of 1000 mg/kg bb showed that three mice undergo degeneration and necrosis, and four mice undergo infiltration of inflammatory cells.
