Kemampuan Ekstrak Jamur Lingzhi dalam Menghambat alpha Glucosidase dan Menurunkan Kadar Gula Darah pada Tikus Hiperglikemia

Abstrak

ABSTRAK Jamur lingzhi telah digunakan secara luas oleh masyarakat secara empiris sebagai pengobatan tradisional untuk menurunkan kadar glukosa darah.Tujuan penelitian ini untuk mengetahui kemampuan ekstrak jamur lingzhi (Ganoderma lucidum) dalam menghambat a-glukosidase pada tikus hiperglikemia.Ekstrak etanol jamur lingzhi diuji daya hambatnya terhadap a-glukosidase menggunakan metode spektrofotometeri.Tikus yang digunakan sejumlah 25 ekor dibagi menjadi lima kelompok. Empat kelompok tikus (P1-P4) diberi sukrosa 80% sebanyak 2 mL secara oral, dan satu kelompok tikus (P0) sebagai kontrol negatif. Sepuluh menit sebelum pemberian sukrosa, tikus kelompok dua (P2), tiga (P3), dan empat (P4), diberi ekstrak jamur lingzhi dengan dosis 10%, 20%, dan 30% sebanyak 1 mL secara oral, sedangkan kelompok satu (P1) sebagai kontrol positif hiperglikemia. Setelah itu,semua kelompok dilakukan pemeriksaan kadar glukosa darah pada menit ke 0, 10, 30, 60, 90, dan 120 seteleh pemberian sukrosa. Hasil penelitian menunjukkan, ekstrak jamur lingzhi mempunyai kemampuan menghambat terhadap a-glukosidase dengan nilai inhibition concentration(IC50) sebesar 162,01 ?gmL-1. Hasil pemeriksaan kadar glukosa darah menunjukkan bahwa pemberian ekstrak jamur lingzhi dengan dosis 30% lebih cepat menurunkan kadar glukosa darah dibandingkan dengan dosis 10% dan 20%. Kata-katakunci :jamur lingzhi,larutan sukrosa, hiperglikemia, IC50 ABSTRACT Lingzhi mushrooms has been widely used by community empirically for the traditional medicine for lowering blood glucose levels. The purpose of this study was to determine the ability of the lingzhi mushroom (Ganoderma lucidum) extract to inhibit á-glukosidase hyperglycemia in rat. Lingzhi mushroom ethanol-extract were tested for their inhibition against á-glucosidase using spectrophotometry method. A total of 25 rats used in this study were divided into five groups. Four groups of rats (P1-P4) were given 2 ml of sucrose 80% orally, and one group of rats (P0) as a negative control. Ten minutes before giving sucrose, three groups (P2-P4) of rats were given the lingzhi mushroom extract of 1 ml orally at a dose of 10%, 20%, and 30% respectively, while the second groups (P1) as a positive control of hyperglycemia. After that in all groups were examined the blood glucose levels at 0, 10, 30, 60, 90, and 120 minute after sucrose administration. The results showed, lingzhi mushroom extract has the ability to inhibit á-glucosidase with IC50 value of 162,01 ìgmL-1. The result of blood glucose test showed that administration of lingzhi mushroom extract at a dose of 30% faster lowering blood glucose levels compared with a dose of 10% and 20%.
