Perkembangan embrio sapi pada media biakan dengan ko-kultur biakan monolayer sel kumulus dan sel epitel tuba fallopii

Abstrak

Abstract Naturally in mammal just following fertilization that occurred in oviduct will be continued the development of embryo to reach early morula in this part of genital tract and of course as long as an environment contributed. This study was aimed to monitoring the ability of two different monolayer cells as well as cumulus and fallopian tube cells to maintain and develop of embryo until 6 days in-vitro. The cumulus and fallopian tube cells was prepared and adjusted to obtain concentration 1.9 x 107 cell/ml and cultured under media TCM 199 with 10 % FCS and 10 % EMS and incubated in incubator with temperature 38.5 0C, 95-97 humidity and 5 % CO2. All the monolayer cells were change the media every 3 days. The embryo having 2-4 cells were transferred to co-culture monolayer either cumulus or fallopian tube cells in which was at day 3 of culturing and continued until 6 days of commencement of embryo transferred or several morula developed. The results showed that quantitatively much more embryo developed to morula (40 %) in co-culture fallopian tube cells compared with only (21.9 %) that reach stadium morula in co-culture cumulus cells. This mean was quite significantly different between both co-culture. That mean embryo more survive in fallopian tube than in cumulus cells is due to product cell secreted from fallopian tube much more complete nutrition and containing less progesterone concentration compared with cumulus cells.
