Abstract Ultraviolet light (UV) can damage the skin and affect the blood so that antioxidants contained in hibiscus leaves are needed. The purpose of this study was to determine the effect of antioxidants in hibiscus leaf simplisia ointment (Hibiscus rosa-sinensis L.) on the hematological profile of white rats (Rattus norvegicus) after UV exposure. The study design used a complete randomized design (CRD) method with a sample of 30 female Wistar strain white rats (100-150 grams) divided into 6 treatments, each group consisting of 5 rats, i.e. (P0) negative control sample without ointment and UV exposure, (P1) positive control sample without ointment and UV exposure only, and (P2, P3, P4, and P5) samples with hibiscus leaf simplisia ointment concentrations of 10%, 20%, 30%, and 40% respectively topically on a shaved back of 16 cm2 , then exposed to UV. Blood specimens were taken one day after treatment through the orbital vein, then a complete hematology test was performed to determine the number of erythrocytes (RBC), hematocrit value (HCT), hemoglobin (Hb) level, and leukocyte count (WBC). The results showed that the administration of hibiscus leaf simplisia ointment can maintain the number of RBC, HCT value, Hb level, and the number of WBC in the treatment group rats (P2, P3, P4, and P5) with hibiscus leaf simplisia concentrations (10%, 20%, 30%, and 40%) showed results that were not significantly different (P>0.05) from the negative control treatment rats (P0). From the results of this study it can be concluded, the administration of hibiscus leaf simplisia ointment to white rats after UV exposure can maintain the number of red blood cells, hematocrit value, hemoglobin level, and leukocyte count.
