PENGARUH PERBEDAAN METODE EKSTRAKSI TERHADAP FINGERPRINT FITOKIMIA HPTLC

Abstrak

The influence of extraction method on the phytochemical chromatographic fingerprint HPTLC Cannabissp. had been conducted. Cannabinoids of leaves, flower, seed, and stem were extracted by maceration and soxhletation method using petroleum eter, methanol, diethyleter, as solvent. Concetrated extracts were spotted HPTLC Si 60 GF254. Plate was eluted with hexane-diethyleter (80:20,v/v) in ascending twin chamber. Each peak was scanned on Camag TLC Scanner III and identification of each in-situ spectra were based on Rf and library spectra. The HPTLC chromatograms were analyzed by using principal component analysis (PCA) and hierarchal cluster analysis (HCA). The aim of this research was to obtain the better extraction method in propose herbal medical standardization and drugs profiling in forensic propose. The extraction method played importance role on the total chromatographic fingerprint of Cannabis sp. This influenced the PCA and HCA results. Maceration Cannabis spwith diethyleter introduced highest recovery of cannabinoids. The highest THC contains and the lowest degradation of THC was obtained from seed. There were no significant contains of major cannabinoids between leaves, flowers and stems.
