Abstrak
ABSTRACT Bali is unique, because the community members are grouped by clan or lineage and they called “soroh” but the genetic evidence not declare yet. Pande clan is a clan that has a wide distribution in Bali. This research was conducted to determine the allele variations of mitochondrial DNA (mtDNA) from Pande Clan in Bali used Polymerase Chain Reaction-Fragment Length Polymorphism method (PCR-RFLP) for forensic database. Samples were taken by purposive sampling consisting of 35 people of Pande clan. DNA from mucosal swab was extracted using phenol-chloroform modified method, DNA amplification of D-loop region of mtDNA, PCR product digestion with five restriction enzymes and the electrophoresis was runed on 1 % of agarose gel and PAGE 10%. The amplification resulted in nine variations of allele. Two types of restriction fragment patterns was found each in digestion with enzyme Hae III, Hinf I, Mbo I dan Dde I . For a while the area mtDNA D-Loop not recognize in enzyme Hind III. The Hind III enzyme is monomorphic, so unuseful to distinguish individual DNA profile.
