IMMUNOMODULATOR POTENCY OF EUPHORBIA MILII AND PROPOLIS COMBINATION TEA (EMP) THROUGH THE SECRESION OF GRANZYME B THAT IS CONNECTED WITH LUNG DAMAGE AND HEPAR TOXICITY IN MYCOBACTERIUM TUBERCULOSIS INFECTED MICE

Abstrak

. Euphorbia milii flower has been shown to increase NKp46 expression of NK cells and the secretion of IL-17. Propolis as a natural immunomodulator can also increase the secretion of IL-17. Granzyme B is a mediator secreted by CD8 T and NK capable of lyseing infected macrophages. The study was aimed to test the immunomodulator potency of E.milii and propolis (EMP) tea through Granzyme B secretion, which was associated with lung damage and hepar toxicity in Mycobacterium tuberculosis (M.tb) infected mice. EMP tea (1: 3) were was prepared at Udayana University, given for 28 days in groups of P1, P2 and P3. Termination of group were done at 24 hours (K1, P1), 48 hours (K2, P2) and 7 days (K3, P3) post M.tb infection. Lung tissue prepared for inspection of Granzyme B due to ELISA procedure (Bt-Lab); lung damage were observed due to HE procedure and Hepar toxicity were tested due to clinical chemistry procedure. From the result we found the highest concentration of Granzyme B in P3 (409.32 ng / l) and the lowest in K1 (316.43 ng / l); there were no lung granuloma formation in all of group and there were normaly hepar function in all group. There was no significant difference between group in granzyme B secretion may due to the duration of EMP tea administration only 28 days did not activate cytotoxic T cell to secretion Granzyme B. There were also a negatif strong correlation between SGOT level with lung damage it may due to decreased SGOT may occur in fibrosis, where increased lung damage can also be caused by fibrosis, whether in this study there were found no fibrosis.
