Uji Proliferasi dan Ekspresi P53 Ekstrak Etanol Minyak Buah Merah terhadap Sel Kanker Serviks SiHa

Abstrak

Backgrounds: The red fruit (Pandanus conoideus Lam.) has been used for cancer treatment empirically. It contains ß-carotene and ?-tocopherol, which are the source of antioxidant. Aim of our study was to investigate anti proliferative effect and tumor supresor gene p53 expression of red fruit oil ethanol extract on cervix cancer cell line SiHa. Methods: Proliferation and tumor supressor gene p53 expression studies were performed in vitro. The study of antiproliferative effect was conducted in six dose variations of red fruit oil ethanol extract and evaluated using tryphan blue staining methods after 24, 48, and 72 hours of incubation. The study of tumor supressor gene p53 expression was performed in three dose variations of red fruit oil ethanol extract and was evaluated using imunocytochemistry staining method after 24 hours of incubation. Results: The result showed that the proportion of proliferation inhibition on cervix cancer cell line SiHa induced by red fruit oil ethanol extract at highest concentration (0.125 ?g/mL) after 24, 48, and 72 hours of incubation were 97.97%; 100.0%; and 100.0%, respectively. If compared with positive control (doxorubicin), the proportion of proliferation inhibition on cervix cancer cell at its highest concentration after 24, 48, and 72 hours of incubation were 85.31%; 94.43%; and 99.82%, respectively. There were significance differences in the proportion of proliferation inhibition among groups in each incubation period (p<0,001). The result also showed that red fruit oil ethanol extract increased tumor supressor gene p53 expression at concentration 0.06250; 0.03125; and 0.01562 ?g/mL by 43.69%; 61.99%; and 18.14%, respectively. If compared with positive control, its p53 expression at three dose variations (3.75; 1.875; 0.9375 ?g/mL) were 42.63%; 41.88%; and 35.94%, respectively. There were significance differences in the p53 expression among groups (p<0,001). Conclusions: In conclusion, red fruit oil ethanol extract has antiproliferative effect and increases tumor suppressor gene p53 expression on cervix cancer cell line SiHa in vitro.
